چکیده

In this work, we have studied the impact of various stages cryopreservation by vitrification in ethylene glycolsucrose medium on plasma membranes adhesive properties 8-cell murine embryos and their specific electrical conductivity. Embryos were divided into following experimental groups: control, group 1 (incubation medium) 2 (complete cycle cryopreservation). The embryo exposure to did not affect ability cell-to-cell adhesion. After a complete cryopreservation, no adhesion was observed. Electrical conductivity determined using pulsed conductometry. incubation medium, resistance electric pulse shown decrease, that manifested phenomenon irreversible breakdown. average values varied within ranges: ((12.1 ± 1.5)...(55.5 2.6)), ((28.7 5.7)...(44.± 8.9)), ((31.0 9.3)...(87.9 26.1)) μS/cm groups 2, respectively. These findings may be explained appearance first structural disorders lipid bilayer even at stage medium. lack blastomeres after vitrification-warming testified damage membrane proteins.

O-6: Cryopreservation and Long-Term Maintenance of Bovine Embryo-Derived Cell Lines

Background: The aim of this study was to develop methods for cryopreservation and long-term maintenance of putative bovine embryonic stem cells (ESCs). Materials and Methods: Putative bovine ESC (bESC) lines (n=3) isolated in conventional medium were used to compare slow-freezing and vitrification. Results: After warming, vitrified cells (96.9%) demonstrated significantly (p<0.05) better surviv...

Mouse embryo cryopreservation utilizing a novel high-capacity vitrification spatula.

Embryo cryopreservation is an indispensable technique in reproductive programs and in animal facilities where genetically modified mice are used extensively. Here we report the use of a vitrification spatula (VS) that can be readily homemade and has a large holding capacity to vitrify preimplantation mammalian embryos in a micro-drop employing ultra-rapid cooling in liquid nitrogen (LN2). Vitri...

Effect of LH Treated Ovine Oviductal Epithelial Cell Co-Culture System on Murine Pre-Embryo Development

Background This study was designed to develop a new co-culture system, assess the effect of luteinizing hormone (LH) using sequential media to promote development and increase the quality of 2-cell murine embryos through the 8-16 cell stage to morula and blastocyst stages. MaterialsAndMethods Monolayers for co-culture were prepared from ovine oviduct epithelial cells (OOEC) in DMEM/F12 medium a...

Biophysical Characteristics of Successful Oilseed Embryo Cryoprotection and Cryopreservation Using Vacuum Infiltration Vitrification: An Innovation in Plant Cell Preservation

Heterogeneity in morphology, physiology and cellular chemistry of plant tissues can compromise successful cryoprotection and cryopreservation. Cryoprotection is a function of exposure time × temperature × permeability for the chosen protectant and diffusion pathway length, as determined by specimen geometry, to provide sufficient dehydration whilst avoiding excessive chemical toxicity. We have ...
